چکیده

Vernalization is one of the most important traits for the adaptability of wheat; detecting variation in genes regulating the response of wheat to vernalization is of importance for classifying winter phenotypes. Ecotilling is a high throughput reverse genetics approach for detecting single nucleotide polymorphisms (SNPs) in the genes of a natural population. To simplify methodology and decrease costs, a modified Ecotilling method based on agarose gel electrophoresis was used to discover the occurrence of SNPs in the wheat vernalization gene VRN-A1 in a natural population of 106 accessions of hexaploid wheat varieties and their association with the winter/spring phenotype. Based on the SNPs detected, these accessions fell into three haplotypes, namely, ́A ́, ́B ́ and ́C ́, each with five, six, and three variations, respectively. The winter phenotype was mainly determined by haplotype ́A ́ and ́B ́, while the spring phenotype was mainly determined by haplotype ́C ́. After sequence confirmation, three SNPs ( ́A/́, in the fourth intron; ́-/A ́, in the sixth intron; ́T/C ́, in the seventh exon) were associated with wheat vernalization and another three mutations ( ́G/T ́ in the second intron; ́T/C ́ in the fourth exon; ́CA/-́, in the fourth intron) had no identified effect. Therefore, Ecotilling with detection of DNA fragments by agarose gel electrophoresis can be used as an efficient method for detecting SNPs in genes of polyploidy materials, such as wheat.

Polymorphism of waxy proteins in Spanish hulled wheats

Hulled wheats are neglected crops that have potential in plant breeding programmes of modern durum and common wheat. Among these wheats, three species were widely cultivated in Spain until the mid 20th century: Triticum monococcum ssp. monococcum (einkorn), Triticum turgidum ssp. dicoccum (emmer) and Triticum aestivum ssp. spelta (spelt). One important aspect of wheat grain quality is starch co...

Heading Date QTL in Winter Wheat (Triticum aestivum L.) Coincide with Major Developmental Genes VERNALIZATION1 and PHOTOPERIOD1

In wheat (Triticum aestivum L.), time from planting to spike emergence is influenced by genes controlling vernalization requirement and photoperiod response. Characterizing the available genetic diversity of known and novel alleles of VERNALIZATION1 (VRN1) and PHOTOPERIOD1 (PPD1) in winter wheat can inform approaches for breeding climate resilient cultivars. This study identified QTL for headin...

Copy Number Variation Affecting the Photoperiod-B1 and Vernalization-A1 Genes Is Associated with Altered Flowering Time in Wheat (Triticum aestivum)

The timing of flowering during the year is an important adaptive character affecting reproductive success in plants and is critical to crop yield. Flowering time has been extensively manipulated in crops such as wheat (Triticum aestivum L.) during domestication, and this enables them to grow productively in a wide range of environments. Several major genes controlling flowering time have been i...
